Persistent immunity after therapeutic vaccination targeting HER2/neu correlated with post-vaccination magnitude of interferon gamma Elispot responses

چکیده
Methods Interferon gamma Elispot data was abstracted from patients enrolled in 3 clinical trials (n = 86) utilizing therapeutic vaccination targeting HER2/neu with long-term follow-up. A positive response/immunity was defined as a precursor frequency of greater than 1:20,000 or 50 interferon gamma corrected spots per well/10 peripheral blood mononuclear cells. Responses were evaluated at baseline study entry, at one month after completion of vaccinations and at long-term follow-up, the last time point available at week 24-36. Correlative statistical analysis was performed using Pearson product-moment correlation coefficients of least-squares regression.

Concurrent trastuzumab and HER2/neu-specific vaccination in patients with metastatic breast cancer.
PURPOSE The primary objectives of this phase I/II study were to evaluate the safety and immunogenicity of combination therapy consisting of concurrent trastuzumab and human epidermal growth factor receptor 2 (HER2)/neu-specific vaccination in patients with HER2/neu-overexpressing metastatic breast cancer. PATIENTS AND METHODS Twenty-two patients with stage IV HER2/neu-positive breast cancer r...
متن کاملEvaluation of Humoral and Cell-mediated Immunity of Two Capripoxvirus Vaccine Strains against Lumpy Skin Disease Virus
Background and Aims: Prevention of Lumpy skin disease (LSD) in cattle is heavily dependent on vaccination. Since the genetic structure of LSD virus has the close relatedness with other Capri-pox virus (CaPV). Therefore, the use of vaccine strains of CaPV derived from sheep and goat would be useful to protect cattle against LSD. Materials and Methods: The susceptible calves of dairy farms were ...
متن کاملEarly gamma interferon and interleukin-2 responses to vaccination predict the late resting memory in malaria-naïve and malaria-exposed individuals.
Two different cell populations respond to potent T-cell-inducing vaccinations. The induction and loss of effector cells can be seen using an ex vivo enzyme-linked immunospot (ELISPOT) assay, but the more durable resting memory response is demonstrable by a cultured ELISPOT assay. The relationship of the early effector response to durable resting memory is incompletely understood. Effector pheno...
متن کاملResults from a phase I clinical study of the novel Ii-Key/HER-2/neu(776-790) hybrid peptide vaccine in patients with prostate cancer.
PURPOSE Active immunotherapy is emerging as a potential therapeutic approach for prostate cancer. We conducted the first phase I trial of an Ii-Key/HER-2/neu(776-790) hybrid peptide vaccine (AE37) with recombinant granulocyte macrophage colony-stimulating factor as adjuvant in patients with HER-2/neu(+) prostate cancer. The primary end points of the study were to evaluate toxicity and monitor p...
متن کاملTargeted delivery of tumor antigens to activated dendritic cells via CD11c molecules induces potent antitumor immunity in mice.
PURPOSE CD11c is an antigen receptor predominantly expressed on dendritic cells (DC), to which antigen targeting has been shown to induce robust antigen-specific immune responses. To facilitate targeted delivery of tumor antigens to DCs, we generated fusion proteins consisting of the extracellular domain of human HER or its rat homologue neu, fused to the single-chain fragment variable specific...
